No Preferences button
I have been using Calibre on a PC for several years without any issues. I am now trying to set it up on a new MacBook Pro. Whilst Calibre appears to download and setup correctly on the MacBook Pro there is no Preferences button on the GUI.

Try widening the window. You can also invoke preferences with Cmd-P. Within Preferences there are options to change the layout and icon size.

On OS X you access preferences from the "calibre" entry in the menubar at the top, which is standard for OS X applications, I believe.
