When you say nothing works it doesn't tell me anything. With no device connected does calibre open. Can you use calibre independent of any device?

These errors are typical of a corrupt file system on your device. reformatting your device and starting fresh might help with these errors. Before you do that ensure your device is backed-up. Contact other Cybook Opus users or the manual to ensure the safe way to reformat your device.

so I did it

thanks on the hint

but now I have another problem: the device doesn't differentiate between SD an device memory- everithing is together, and it doesn't show the folder structure anymore...

when I connect the device to the computer, I can see the folders, but if I want to browse through the device (once it is disconnected), it doesn't show any

A semi-connected note: The Bookeen devices seem to be rather finicky about filesystem integrity. You need to make sure that you safely disconnect the device (use the eject feature your OS provides). If they get corrupted, scan the disk(s) for errors and let the scan program fix those.
