Do you still have the original card? The first thing to do would be to see if the reader still works with it. If it does, you could clone it onto your 16gb card again.
If not, have you tried a Reset (push a paperclip into the reset hole)?
If that fails, then (unless someone else has a better idea) you'll need to try a factory reset which will try to rebuild the card. Note that this will remove any books that you have on the card, but you should be able to install them again afterwards.
