The /opt/amazon/ebook/prefs/com.amazon.ebook.booklet.reader/reader.pref.default suggests it's a file on the Kindle - one of the Kindle's own system files, a settings file. It's surely just a text file, editable with a text editor. You'd need access to the Kindle's filesystem;
